Boris Johnson told Vladimir Putin that expanding into Ukraine would be a "tragic error" during their phone chat, which had been delayed due to the 'partygate' controversy.
The prime minister is said to have expressed his 'deep concern' as an estimated 100,000 Russian troops gather on the Ukrainian border with war feared to be 'imminent'.
Downing Street said he told the president Ukraine has a right to want Nato membership – as Mr Putin demands legally binding guarantees from the US and Nato that the country will never join the bloc.
The 45-minute call was finally held on Wednesday afternoon after the Tory leader postponed it in order to deal with the fallout of the Sue Gray report.
In a statement today, a No 10 spokeswoman said: 'The Prime Minister expressed his deep concern about Russia's current hostile activity on the Ukrainian border.
'He emphasised the need to find a way forward which respects both Ukraine's territorial integrity and right to self-defence.
'The Prime Minister stressed that any further Russian incursion into Ukrainian territory would be a tragic miscalculation.'

Mr Johnson was said to have stressed that all European democracies have the right to aspire to Nato membership, and that the right 'fully applies' to Ukraine.
The call has overall been characterised as constructive and thorough, with the two world leaders welcoming communication on issues including the climate crisis.
'They agreed to apply this spirit of dialogue to the current tensions in order to find a peaceful resolution', the spokeswoman added.
